THE RESPONSE OF THE CENTRAL GHRELIN SYSTEM TO THE DEPRIVATION OF CUBS RATS FROM THE MOTHER IN EARLY ONTOGENESIS
Abstract
Introduction. Early-life stress can exert profound and long-lasting effects on organism development, manifesting as impaired adaptation, increased anxiety, depressive behavior, and other emotional disturbances. The aim of this study was to investigate the response of the central ghrelin system to maternal deprivation during early ontogeny in Wistar rats. Materials and methods. The experiment involved 40 male Wistar rats (6 litters), which were randomly divided into two groups: control (Con, n=20) and maternal deprivation (MD, n=20). From postnatal day 2 to day 12, pups in the MD group were separated from their mothers for 180 minutes daily and placed in individual plastic containers for ten consecutive days. At the age of 90–100 days (body weight 200–250 g), the animals were sacrificed by decapitation. The hypothalamus, amygdala, and prefrontal cortex were rapidly dissected, immediately frozen in liquid nitrogen, and stored at temperature–80 °C until RT-PCR analysis. Results. No statistically significant differences were found in ghrelin receptor mRNA expression between groups in the hypothalamus or prefrontal cortex. However, in the amygdala, the MD group showed a significant increase in Ghsr1a gene expression compared to the control group. Conclusion. These findings suggest that maternal deprivation predominantly affects the extrahypothalamic regulation of the ghrelin system in the brains of the studied animals.
