Experimental modeling of autoimmune hepatitis

Abstract

Introduction. Autoimmune hepatitis (AIH) is a chronic inflammatory liver disease characterized by the presence of specific autoantibodies and elevated gamma-globulin levels targeting hepatocytes. Currently, it is recognized as a widespread autoimmune disorder with challenges in early diagnosis. The global annual incidence rate is estimated at 1.37 per 100,000 population. Aim — to create an experimental model of autoimmune hepatitis in laboratory animals. Materials and methods. Male Wistar rats (n=5) received an oral solution of sodium silicate once a week for five weeks. After the experiment, biochemical and histological analyses of liver tissue were performed. Results. Compared with the control group, the experimental rats showed significant increases in intracellular enzyme levels: AST (137.6±8.8 U/L) and ALT (35.2±2.2 U/L) (p <0.05), confirming hepatocyte damage. Alkaline phosphatase (ALP) increased significantly (28.9±2.3 U/L), indicating the development of cholestatic syndrome (p <0.05). Total cholesterol (1.9±0.2 mmol/L) and triglycerides (0.7±0.2 mmol/L) were elevated (p <0.001). Total bilirubin rose to 12.5±0.9 µmol/L. Histological examination revealed hepatocyte damage and inflammatory infiltration consisting of lymphocytes and plasma cells. Conclusion. An experimental model of autoimmune hepatitis was successfully established. Characteristic histological and biochemical changes were identified. These findings may serve as a foundation for future research on autoimmune hepatitis.
